Product Overview
The Omega Bio-tek E.Z.N.A.® Viral RNA Kit is a manual, silica-based spin-column system for purification of viral RNA and DNA from cell-free biological fluids. Primary sample types include plasma, serum, urine and cell culture supernatant.
The workflow uses the reversible nucleic-acid binding properties of the HiBind® RNA Mini Column. Samples are lysed under denaturing conditions intended to inactivate RNases and protect viral RNA from degradation. After binding to the column membrane, contaminants are removed through sequential wash steps and purified nucleic acids are eluted in nuclease-free water.
The kit supports either centrifugation or vacuum processing and does not require an organic extraction. Purified nucleic acids are suitable for downstream research applications such as qPCR and RT-qPCR when used according to the applicable protocol.
For Research Use Only. Not for use in diagnostic procedures.
150 µL Sample Input
The standard protocol begins with 150 µL of cell-free sample.
20–50 µL Elution
Purified nucleic acids are eluted in a low volume of nuclease-free water.
1–24 Samples
Manual processing by centrifugation or a compatible vacuum manifold.

Viral Nucleic Acid Purification Workflow
1. Lyse
Combine the sample with QVL Lysis Buffer and carrier RNA under the conditions specified in the protocol.
2. Bind
Add ethanol and transfer the prepared lysate to a HiBind® RNA Mini Column.
3. Wash
Wash the bound nucleic acids with VHB Buffer and RNA Wash Buffer II.
4. Elute
Elute the purified nucleic acids in 20–50 µL of nuclease-free water.
This summary does not replace the complete manufacturer protocol. Follow the current manual for reagent preparation, volumes, incubation, centrifugation or vacuum settings and storage.

Important Product Information
- This product is for research use only and is not intended for diagnostic procedures.
- The kit does not selectively separate viral RNA from cellular RNA or DNA. If cellular nucleic acids are present, they may also be purified.
- Acellular body fluids are recommended when isolation of viral RNA without cellular nucleic acids is required.
- Sample compatibility, handling, pretreatment and storage must follow the current manufacturer protocol and the laboratory’s validated procedures.
- Carrier RNA requires separate preparation and storage. RNA Wash Buffer II and VHB Buffer require ethanol to be added before use.
- The linked Jant-hosted manual is Revision 4.0 dated September 2019. Contact Jant Pharmacal to confirm whether a newer revision is applicable before beginning work.

Frequently Asked Questions
The current manufacturer product page describes the kit as isolating viral RNA and DNA. The method is not selective for viral RNA when other cellular RNA or DNA is present in the sample.
The primary starting materials are cell-free fluids such as plasma, serum, urine and cell culture supernatant. Confirm any additional sample matrix against current Omega Bio-tek application guidance before use.
The standard protocol uses 150 µL of plasma, acellular body fluid, urine or cell culture supernatant.
The protocol calls for elution in 20–50 µL of nuclease-free water.
Yes. The manual provides both centrifugation and vacuum protocols. Vacuum processing requires a compatible manifold, flask, tubing and vacuum source supplied by the laboratory.
No. The E.Z.N.A. Viral RNA Kit uses silica-membrane spin-column purification and does not require an organic extraction.
The manufacturer identifies qPCR, RT-qPCR and related downstream research applications. The laboratory must determine suitability and validate the complete workflow for its intended research use.
No. The laboratory must supply 100% ethanol. Ethanol is added to RNA Wash Buffer II and VHB Buffer according to the preparation instructions for the selected kit size.
The available configurations are R6874-00 for 5 preparations, R6874-01 for 50 preparations and R6874-02 for 200 preparations.
