Accutest Standard Respiratory RT-PCR Panels
Accutest Respiratory RT-PCR Panels provide a comprehensive, multiplexed real-time PCR solution for the detection of high-prevalence respiratory pathogens associated with upper and lower respiratory tract infections. Each panel includes optimized primers and probes for a broad range of clinically relevant viral and bacterial organisms — including influenza A/B, RSV, SARS-CoV-2, human metapneumovirus, and common community-acquired bacterial agents such as Streptococcus pyogenes and Haemophilus influenzae. An internal control (Rnase P / RPP30) is incorporated in every reaction to validate nucleic acid extraction, amplification, and detection, supporting consistent, reliable results.
Panels are available in multiple configurations — full 384-well plates, 96-well plates, and tube format (100 reactions) — allowing laboratories to match throughput, workflow, and instrument capacity without modifying assay content. MasterMix is available separately for one-step multiplex RT-PCR.
By consolidating multiple respiratory pathogen targets into streamlined multiplex reactions, the Accutest Respiratory RT-PCR Panels reduce turnaround time and hands-on labor compared to culture-based or singleplex molecular methods, conserve reagents, and improve operational efficiency. Pre-spotted plate formats eliminate manual primer and probe preparation, helping reduce contamination risk and pipetting variability.
All products are shipped on dry ice and stored at –15 °C to –25 °C for stability and consistency across lots. Typical reaction setup supports 10 μL final volume in 384-well systems (or 20 μL in 96-well systems), with both standard and fast cycling protocols compatible with common real-time PCR instruments.
For research use only (RUO); not validated for diagnostic procedures. The U.S. CDC reports that respiratory pathogens including influenza, RSV, and parainfluenza remain major contributors to seasonal illness (CDC Respiratory Virus Surveillance).
Multiplex RT-PCR for Respiratory Infection Research
The Accutest Standard Respiratory RT-PCR Panels are designed to detect a wide range of viral and bacterial agents commonly associated with respiratory illness. Targets include:
- Common respiratory viruses: Influenza A, Influenza B, RSV A/B, seasonal human coronaviruses (229E, NL63, OC43, HKU1), SARS-CoV-2, human metapneumovirus, parainfluenza viruses 1–4, adenoviruses, rhinovirus, and enterovirus D.
- Atypical / intracellular pathogens: Mycoplasma pneumoniae and Chlamydia pneumoniae, organisms frequently associated with atypical or community-acquired pneumonia.
- Typical/community-associated bacteria: Haemophilus influenzae, Moraxella catarrhalis, Streptococcus pyogenes, Staphylococcus aureus, Legionella pneumophila, and related antimicrobial resistance markers (e.g. mecA).
Multiple catalog configurations are offered to accommodate diverse research workflows and experimental throughput requirements.
MasterMix
Broad-Range 4X MasterMix is a versatile one-step RT-PCR solution optimized for multiplex respiratory applications, offering fast, reliable performance and seamless compatibility with all Accutest RT-PCR panels.
Composition and Features
- Integrated enzymes: Hot-start reverse transcriptase and hot-start DNA polymerase support efficient cDNA synthesis and amplification of RNA viruses such as influenza, RSV, SARS-CoV-2, and seasonal coronaviruses.
- Balanced reaction mix: Includes optimized dNTPs, MgCl₂, proprietary enhancers, and stabilizers to maximize performance under high-plex respiratory conditions.
- One-step RT-PCR: Built-in reverse transcription streamlines detection of single-stranded RNA viruses in a single reaction.
Advantages
- High performance across multiplex respiratory RT-PCR panels
- Reliable amplification from challenging respiratory specimen types
- Reduced assay time with integrated reverse transcription
- Universal compatibility with Accutest RT-PCR panel designs
- Flexible use across high-throughput and low-throughput workflows
Workflow Overview
The Accutest Standard Respiratory RT-PCR Panels are designed for ease of use and consistency:
- Add MasterMix to the wells or tubes containing pre-spotted primers and probes.
- Introduce direct specimen or purified nucleic acid template.
- Seal, mix, and briefly centrifuge the plate.
- Run the cycling program on a compatible real-time PCR instrument.
The recommended reaction volume is 10 μL for 384 systems and 20 μL for 96 systems. Both standard and fast cycling protocols are supported.
